Substrate-controlled Enzymatic Glycosylation for Complex Oligosaccharide Synthesis
My laboratory focuses on the synthesis of human milk oligosaccharides (HMOs). The use of enzymatic strategies for glycan synthesis not only provides better control of stereoselectivity but also avoids multistep synthetic manipulations required by chemical approaches to complex carbohydrate synthesis. These biologically important HMOs will serve as essential probe molecules and/or nutrient resources to explore the interaction of HMOs with human gut microbiota. This work has applications in vaccine development and drug discovery. A major contribution from my lab is the development of a sequential one-pot enzymatic synthesis system using a panel of glycosyltransferases (GTs) and sugar-nucleotides generating enzymes, which has allowed the facile preparation of a library of HMOs (~100 glycans). This is one of the largest collections of this type in the work and importantly we have made these compounds in quantities that facilitate a large number of biological studies.
